One of the cationic colouring (dimethyl-metilene blue) techniques used in Clinical Laboratories for the screening of mucopolisaccharidosis recommends the GAG or mucopolisaccharides precipitation with CPC In this paper was introduced a modification that consisted in eliminating the precipitation phase in order to avoid any posible loss of sample which could take place in the consecutive decantations of the urines; moreover, it leads to a simplification of the technique. The results confirmed the initial hypothesis; a significant diference (p<0.01) between the measured urinary GAG in two confirmed mucopolysccharidosis (the highest levels) and a normal adult (the lowest level) was found. Within the medium levels, both techniques (precipitation and no precipitation) have identical results. In conclusion, the suggested modification not only simplifies the technique but also it makes it more reliable in the extreme urinary GAG levels.
