Oncological processes are characterized by an uncontrolled cell replication always associated to DNA implication. DNA molecule is the biological target for the action of many antitumor drugs and potential antineoplastic compounds. As a result of many years of work, researchers have been able to design and synthesize a great number of compounds binding to DNA through different mechanisms, among which many known antitumor agents are found. Several techniques allow us nowadays to establish the type of interactions between DNA and its ligands, some classical and some recently developed. A reasoned combination of methods can help to differentiate the molecular mechanisms of the binding processes, to establish the structure-activity relationships, and, what is more important, to develop reasoned designs for the discovery of new agents.
